This subproject is one of many research subprojects utilizing theresources provided by a Center grant funded by NIH/NCRR. The subproject andinvestigator (PI) may have received primary funding from another NIH source,and thus could be represented in other CRISP entries. The institution listed isfor the Center, which is not necessarily the institution for the investigator.In this study, the PI will examine features of coagulation that lead to bleeding or thrombosis. She will examine procoagulant activity of blood cells. She will perform these studies using a TF-initiated model system of coagulation that utilized stimulated monoctyes. freshly isolated platelets and purified coagulation proteins and correlate parameters of thrombin generation, platelet activation and fibrin clot structure. Five to 50 ml of blood will be drawn from a vein in the subject's arm. Monocytes and platelets will be isolated from the blood and used in subsequent in vitro experiments. The activity of monocytes, platelet activation and thrombin generation under a variety of conditions in vitro will be examined.
