Tuberculosis (TB) takes longer to cure than almost any other bacterial infection. Subpopulations of Mycobacterium tuberculosis (Mtb) become phenotypically drug-tolerant in association with host-imposed stresses that put Mtb into a slowly-replicating or non-replicating state (here called NR for simplicity). These studies will address two challenges: to characterize one of the persistent, metabolically diverse Mtb populations of most importance for TB treatment duration and relapse, and to identify early markers of bacterial clearance in response to chemotherapy. This will advance our understanding of Mtb's persistence and help find drugs that can overcome it.
