This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Structural studies of adeno-associated virus 4: A fairly complete diffraction data set (75.9% complete to 3.2A resolution) were collected for adeno-associated virus serotype 4 crystals, under cryo conditions. The images were indexed with DENZO (1) and scaled and reduced with SCALEPACK. The crystals belong to the orthorhombic crystal system with cell parameters of a=339.6, b=319.2 and c=285.0 A. The virus particle orientation(s) and positions in the crystal unit cell have been determined and data has been phased using the available structure of the homologous feline panleukopenia virus as an initial model. Structure determination and refinement are underway. The production, purification and preliminary X-ray crystallographic studies of the AAV4 particles have been published.
