Research: The proposed research is a systematic study of the partial duplication of the M-bcr that is associated with the t(9;22) in patients with CML.
The specific aims are two fold: 1) determine the frequency, extent and location of the M-bcr duplication with relationship to the M- bcr breakpoint locations on the Philadelphia and 9q+ chromosomes in patients with CML and 2) determine whether the duplication is inherited or acquired. It is anticipated that the results of this study will indicate whether the duplication event occurs during or precedes or succeeds the translocation event. This will provide important insight into the mechanism of the recombinant event and, perhaps, oncogenesis. In addition, this data will minimize the confounding effect that the M- bcr duplication has on breakpoint restriction mapping and the molecular diagnosis of CML.
