The institution listed is for the Center, which is not necessarily the institution for the investigator. Aging is associated with decreased Growth Hormone (GH) secretion, known as somatopause. While in older men this may be in part secondary to diminished secretion of Growth Hormone Releasing Hormone (GHRH), this is not true for women. Ghrelin, a recently discovered gastric peptide, strongly stimulates GH release and its levels are three times higher in young women at the high estrogen phase of the menstrual cycle than in young men. We will examine whether somatopause is associated with a decrease in ghrelin secretion and how the estrogen milieu affects ghrelin secretion. Forty-eight healthy individuals in four age and gender specific groups will be studied. Men will be admitted to the General Clinic Research Center once and women twice during artificially induced 'estrogen rich' and 'poor' periods. During each admission, blood will be drawn for plasma levels of GH and ghrelin for twenty-four hours. After this period, an intravenous dose of GHRP-2, a synthetic substance similar to ghrelin, will be given and GH will be measured to help distinguish whether changes in ghrelin sensitivity contribute to somatopause. Finally, an IVGTT, a test involving intravenous injection of glucose and insulin to assess sensitivity to insulin, will be performed.
