Androgen deprivation therapy (ADT) with orchiectomy or administration of LHRH agonists is standard treatment for metastatic prostate cancer (PC). Accelerated osteoporosis is one of the side effects of the resulting hypogonadism. Pamidronate disodium is a bisphos-phonate that is an inhibitor of osteoclast-induced bone resorption. Pamidronate has been shown to preserve bone mass in women with post-menopausal osteoporosis and in patients with glucocorticoid-induced osteoporosis. We propose a study of men with PC on ADT who will be randomized to receive either pamidronate 90 mg IV or placebo NaCl every 3 months for 1 year. Bone mineral density and biochemical markers of bone turnover will be measured every 3 months during this period and comparisons of these indicators of osteoporosis will be made between the two treatment groups.
