This subproject is one of many research subprojects utilizing theresources provided by a Center grant funded by NIH/NCRR. The subproject andinvestigator (PI) may have received primary funding from another NIH source,and thus could be represented in other CRISP entries. The institution listed isfor the Center, which is not necessarily the institution for the investigator.There has been a rapid rise in the availability and use of unregulated dietary supplements by the general public and also by elite athletes. The extent of use of these products within the sports community is largely unknown. However, it has been demonstrated that many of these products contain androgenic-anabolic steroids (AAS), steroid prohormones (precursors), or steroid-like compounds. The unsuspected oral ingestion of these compounds is a growing concern in the sports community because individuals who consume health supplements may produce urine that is 'positive' upon drug testing. Also, the typical urine profiles of metabolites produced as a result of these exposures has not been studied and it is unknown how this might affect the testing process.The goal of this proposal is to characterize the unique and dynamic human urinary steroid metabolite profile(s) after exposure to selected non-physiologic steroids and prohormones administered as: 1) a single dose, and 2) a maximum of five days of single doses. No long term administration of these products will be undertaken. To achieve this goal, this project has the following objectives:1. Determine the unique profile and time course of elimination of metabolites after single-dose administration of several non-physiologic steroids and prohormones sold in selected dietary supplements.2. Determine the unique profile and time course of elimination of metabolites after multiple-dose administration (5 days) of several non-physiologic steroids and prohormones sold in selected dietary supplements.3. Compare the urine metabolite profiles and time course of elimination that results from exposure to different formulations of several non-physiologic steroids and prohormones sold in selected dietary supplements. The hypotheses for these specific aims are:1. There will be no change in endogenous urine steroid profile after administration of selected steroid prohormones and precursors found in selected dietary supplements.2. There will be no change in the testosterone/epitestosterone ration after administration of selected steroid prohormones and precursors found in selected dietary supplements.
