Background: Many metabolic studies have relied upon retrieving multiple muscle samples from a common site. Recent studies have suggested that obtaining multiple samples from a common site may not be reliable because of potential inflammation from the prior biopsy. Hypothesis: Muscle biopsies taken from a common site will not differ significantly from muscle biopsies taken from different sites and different legs.
Specific Aims and Procedures (summary): Six muscle samples total will be taken from 4 sites total on both legs. We will compare measures from single-biopsy sites with measures from multiple-biopsy sites Experimental Design (summary): We will study 25 subjects both male and female within the age range of 18-40 years. We will infuse stable isotopes of amino acids for 10 hours (0600-1600) to measure the uptake of the isotope into the target blood and muscle samples. Blood samples will be taken at specific time points to assure that adequate levels of isotope are present. Muscle samples will be taken at 3hrs, 6hrs and 9hrs after beginning the infusion of isotope. Subjects will arrive shortly before study begins and will be discharged shortly after the final biopsy is obtained. Significance (summary): A scientifically valid method for acquiring multiple muscle samples while minimizing risks to subjects will serve to ensure data reliability and enhance subject safety.
