This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. This study will test the hypothesis that treating mild asthmatics with rhumAb-E25 reduced Fc&RI expression and signaling activity in lung mast cells as it does in basophils and to determine if IgE removal impairs basophil Fc&RI stability and Fc&RI-kinase coupling. a. To measure effects of rhumAb25 treatment and withdrawal on Fc&RI levels and Fc&RI-mediated degranulation in the lung mast cells and blood basophils of mild asthmatics. b. To explore effects of IgE removal on the stability of Fc&RI subunit interactions and on Fc&RI-kinase coupling in basophils.
