This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The overall goal of the study is to identify the changes in metabolism specifically related to the pre-diabetic state in subjects with positive family history when compared to metabolism associated with the insulin resistant state alone, in the absence of association to the development of disease. Insulin secretion and sensitivity of insulin resistant cohorts will be compared to individuals with and without family history of type 2 diabetes with high insulin sensitivity and high glucose effectiveness. The physiologic changes in metabolism will be related to the functional changes in proteins involved in glucose metabolism and to the genomic mutations specific to the disease.
