This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. We previously developed a continuous inhalation method that measures the overall dose of O3 to an intact human lung, and a bolus inhalation method that characterizes the relative uptake of O3 into different regions of the respiratory tract. In the current student, we will apply these methods to healthy smokers and nonsmokers to determine how smoking influences O3 uptake into the conducting airways compared to O3 uptake into the alveolated airspaces. The physiological responses of the conducting airways and respiratory airspaces will also be measured so that regional dose-response relationships can be determined. We will utilize 30 healthy smokers and 30 healthy nonsmokers who each participate in three research sessions. During session I, the distribution of O3 uptake will be measured by the bolus inhalation method while employing a standard breathing pattern. During session II, overall O3 uptake will be measured on a breath-by-breath basis while a subject continuously breathes 0.30 ppm O3 while exercising on a cycle ergometer. During session III, intended to be a control study, each subject will exercise for one hour while breathing clean air. Physiological responses in sessions II and III will be determined from decrements in FEV1 as well as from the peripheral airspace area (AP) obtained from a CO2 expirogram.
