Specific Aims : This series of experiments focuses on understanding the balance between dilation and constriction in the popliteal artery, a blood vessel that runs behind the knee. We will examine the blood flow and diameter changes in this artery during conditions of increased blood flow, an ice-water bath to stimulate the sympathetic nervous system, and a combination of the two. We will be studying 3 groups of women: ages 20-30, ages 60-79, and older women who are currently on estrogen-replacement therapy as estrogen may influence the responses of blood vessels. Hypotheses: We expect that older women who are NOT taking estrogen will have less dilation and blood flow in the popliteal artery than young. These same older women will also have MORE constriction in the popliteal artery during ice-water stimulus. When the two conditions are examined together, the balance of dilation and constriction will be changed in older women so that they have a greater reduction in dilation than younger women. However, we expect that older women taking estrogen will have arterial responses that are similar to young women! We expect this research will yield information about the regulation of dilation and constriction in the popliteal artery. Declines in submaximal leg blood flow may affect the ability of women to exercise and perform daily activities of living. Knowing how and why blood vessel dilator and constrictor responses change with age and estrogen status in women will help us develop strategies to help treat and prevent altered leg blood flow regulation.
