This subproject is one of many research subprojects utilizing theresources provided by a Center grant funded by NIH/NCRR. The subproject andinvestigator (PI) may have received primary funding from another NIH source,and thus could be represented in other CRISP entries. The institution listed isfor the Center, which is not necessarily the institution for the investigator. There are almost no data on racial differences in oxidative stress, and on racial differences in the effects of cigarette-smoke induced oxidative stress. Among smokers, we hypothesize that blacks might be exposed to substantially higher levels of cigarette smoke free radicals since they are exposed to higher levels of other cigarette smoke metabolites than whites per cigarette smoked. In our previous studies, we determined glutathiolated proteins (GSSP) levels in black smokers, white smokers, and white nonsmokers. Higher GSSP levels were observed in black vs. white smokers, but we did not have the opportunity to conduct GSSP determinations in nonsmoking blacks. To fully understand racial differences in smoke-induced oxidative stress, it is necessary to have a baseline comparison group of both white and black nonsmokers. We thus propose to recruit nonsmoking black subjects through the GCRC. In addition to measuring blood GSSP, we will examine the effects of single nucleotide polymorphisms in oxidative stress genes to determine their impact on these relationships.
