This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. We propose to comprehensively examine the influence of caloric restriction (weight loss) and exercise training (with and without weight loss) on hepatic and peripheral insulin resistance in persons with IGT during a 24 week, randomized, controlled trial. We will utilize a somatostatin, multi-stage, euglycemic clamp technique to specifically examine hepatic and peripheral insulin action. Overall, these studies will be the first to systematically identify the influence of diet and exercise on the pathogenic factors (hepatic and peripheral insulin resistance) that contribute to Type 2 diabetes and CVD
