Morrow, M.D., Co-Director Abstract: The Mass Spectrometry and Proteomics Core (MSPC) provides analytical instrumentation and expertise to serve both the small molecule and proteomic needs of this PPG. The MSPC operates within the Mass Spectrometry Research Center, which houses all MS instrumentation at Vanderbilt, and the Eicosanoid Core Laboratory (directed by Jason D. Morrow M.D., Professor of Medicine and Pharmacology), which provides the technical resources for the preparation of biological samples for the analysis of isoprostanes (IsoPs). The primary functions of the lipid analysis component of this core are to 1) quantify IsoPs for projects in this application and 2) provide instrumentation for the analysis of various lipid oxidation products for investigators in this PPG. Analysis of IsoPs is performed by negative ion chemical ionization (NICI) gas chromatography (GC/MS). LC-MS-MS methods will be applied to the analysis of oxidized lipid mediators. One of these that will be extensively employed in Projects 1 and 2 is coordination ion-spray massSpectrometry (CIS/MS), which is extremely useful for the analysis of complex peroxide mixtures. The Proteomics component of the MSPC provides analytical proteomics services to investigators in the PPG. The major proteomics focus areas in this PPG are the identification of proteins in complex mixtures and the analysis of protein modifications by reactive electrophiles. However, the proteomics component of the MSPC provides a broad range of proteomics-related analyses. Analyses of posttranslational modifications and chemical adducts are done by LC-MS-MS and MALDI-TOF-TOF analyses of peptides from protein digests. Multidimensional LC-MS-MS is used to improve detection of low-abundance modified peptide forms. Novel data mining software can be used to identify unanticipated electrophile adducts from MS-MS data.
