This subproject is one of many research subprojects utilizing theresources provided by a Center grant funded by NIH/NCRR. The subproject andinvestigator (PI) may have received primary funding from another NIH source,and thus could be represented in other CRISP entries. The institution listed isfor the Center, which is not necessarily the institution for the investigator.The objective of this pilot project is to describe the distributionof cytochrome P450 polymorphisms in the Puerto Rican population, in partuclar CYP2D6 and CYP2C19. The P450 family of mixed function oxidases is responsible for metabolizing drugs used in psychiatry. The 2D6 subgroup is highly polymorphic with enxyme activity varying from none to several fold more active than the average. Knowledge of P450 polymorphism helps clinicians to design drug treatment regimens. The distribution of polymorphisms has been determined for Asian, European and African populations, but not for Latin Americans, with the exception of one small study of Mexican Americans. Analysis of P450 polymorphisms in Puerto Ricans will assist prescribing physicians and will contriubte to understanding of P450 genes. We will analyze blood samples of selected subjects using the Roche AmpoliChip mircoarray with the Affymetrix Plate reader and software. Dr. Jose de Leon from the University of Kentucky , a renowned expert in the area, has agreed to assist us in establishing the analysis and to collaborate on the interpretation of the results.
