The institution listed isfor the Center, which is not necessarily the institution for the investigator.Regarding AIDS:The flow cytometry core is a resource to scientists. A research investigator contacts me with an experimental question and together we devise the methodology to address the question. Most protocols must be tailored to cell type, physiology, proliferation rate, and sensitivity to fixative or permeabilization agents. Additionally, we must take into account the nature of the molecule to be studied. We will address specific questions regarding soluble molecules, such as calcium during signal transduction, surface markers which identify the population of interest, and molecules in the cytoplasm of cells. Further, I guide the investigator to prepare the appropriate controls for proper interpretation of the experimental data generated. The number of markers used to quarry the cells can be quite numerous and therefore demand stringent methods for eliminating misinterpretation of the data.Once the experimental question and conditions are defined, the investigator returns to the lab. I will set up templates for each experiment and in doing so train the investigator at the instrument to run their samples as well as read the data collected. Often it takes several settings at the instrument for an investigator to become comfortable with running their samples. Additionally, it will take several more consultations for the investigator to become comfortable with reading and interpreting their data. The majority of my time is spent in the lab training investigators and/or running samples. Performing analysis and preparing figures and/or tables for publications, is another service I offer investigators.
