This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. This Core provides histology services to COBRE and other MMCRI investigators. The Core performs tissue fixation, embedding, and sectioning of frozen, paraffin or plastic embedded specimens. The Core performs histological stains including Hematoxylin-Eosin, Trichrome, von Kossa, Orcein, van Gieson (and others as needed). Immunohistochemical staining with a variety of antibodies is also part of Core services. The Core is operated by a full time histologist with more than 25 years of experience in the field (Kathleen Carrier). The Core director (Volkhard Lindner) oversees Core operations and assists in the interpretation and evaluation of histology specimens. The Core uses the following equipment: Microm HM505E Cryostat for frozen sections, A Microm HM355S and Microm HM325 microtome for paraffin and plastic sections, A Fisher HistoCenter embedding station, Lab-Line Vacuum oven Model 3618 and VWR vacuum oven model 1410 for paraffin infiltration, Slide Warmers and water baths. For to the centralized histology operation an automated tissue processor (Leica TP1020) was brought on line in 2004 through funds provided by this COBRE award. An automated slide stainer (Leica Autostainer XL) and cover-slipping station (Hyperclean) were brought online in 2005. Kathleen Carrier performs all day-to-day operations including tissue processing (Leica TP1020) & special stains; immunohistochemistry; request forms; ordering and maintenance records; select protocol development; and confocal microscopy. Also, on-line are a HM505E Cryostat, HM355S and HM325 microtomes, HistoCenter embedding station, and ovens for paraffin infiltration.
