This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Our tomography graphical user interface, called eTomo, has now reached the significant milestone of inclusion in the IMOD software package. Three beta-releases provided valuable feedback from both local users and those attending our tomography workshop. ETomo is now the primary interface for generating tomograms in the IMOD documentation. This has resulted in numerous user comments expressing appreciation for the greatly improved accessibility to the IMOD package and electron tomography in general. Several areas of new functionality have also been incorporated into eTomo. To address the problem of increased numbers of x-ray counts that occur with directly couple camera systems, we have implemented an automated x-ray removal interface into the pre-processing stage of eTomo. There has also been a need, especially with cryo-tomography, for a trial tomogram generation process that provides coarse tomograms in a short amount of time. We have implemented this functionality as an optional step in the tomogram calculation step. Finally, we have implemented an interface for tomogram trimming and scaling and a file management interface for handling intermediate files. We are now improving the process flow flexibility within eTomo. As currently practiced, sectioned cryo-tomography does not allow for gold fiducials to assist in the fine alignment process. This lack of fiducials as alignment markers requires an alternate data processing path. This path has been designed and is now being implemented. While we are adding the process flow flexibility for non-fiducial alignments we have also chosen to implement the process stream for montaged tomograms. With the addition of these new capabilities, eTomo will provide graphical user interface functionality for most of the electron tomography protocols.
