These molecules contain an N-terminal Ser/Thr kinase domain and a C-terminal domain called the Polo-box domain (PBD). The PBD inhibits the kinase domain in the inactive state but targets the kinase to its substrates and releases the kinase inhibition in the active state. The molecular arrangement of the PBD and the kinase domain, and the mechaism of kinase regulation by the PBD is unknown.
