This subproject is one of many research subprojects utilizing theresources provided by a Center grant funded by NIH/NCRR. The subproject andinvestigator (PI) may have received primary funding from another NIH source,and thus could be represented in other CRISP entries. The institution listed isfor the Center, which is not necessarily the institution for the investigator.Wensel's lab is using cryo-EM to determine the structure of the ion channel TRPV1. This channel plays an important role in the sensation of painful heat and is an important drug target. This structure will likely be the first one determined for a large class of related TRP channels, which play a wide range of roles in sensation and disease. The channel is thought to be a tetramer, but no structural studies have been carried out to date because of a lack of suitable purification methodology. Dr. Moiseenkova has developed a yeast expression system and affinity purification procedure that allows sufficient homogeneous channel to be purified for electron microscopy.
