The institution listed isfor the Center, which is not necessarily the institution for the investigator.The investigation of light-driven signal generation/transduction in light-harvesting protein systems of photosynthetic lower eukaryotes and bacteria is of a wide biological interest. Major progress has been made in understanding the mechanism of generation of an initial signal. However, the main question still remains unanswered: what are the pathways of signal transduction in response to such environmental stimuli as light? In this proposal we address the problem of the molecular mechanism of light-driven signal transduction in light-sensing protein systems. As a representative system we have chosen Avena sativa (oat) phototropin1 (phot1). Phot1 is a plasma membrane-associated flavoprotein that functions as the primary photoreceptor mediating phototropic plant movement, phototropism. Phot1 is composed of two N-terminal light, oxygen, voltage (LOV) sensor domains, denoted LOV1 and LOV2, and an effector C-terminal Ser/Thr kinase domain. The molecular mechanism of signal transduction in phot1 may reveal how the system controls phototropism at the molecular level. Moreover, the project offers the opportunity to compare different folds of LOV domain proteins and reveal common aspects of their signal transduction mechanisms. That is, we can highlight an evolutionarily-conserved pathway of long-range structural connectivity leading from the LOV domain(s) to the effector domain(s) or protein-interaction partners.
