This subproject is one of many research subprojects utilizing theresources provided by a Center grant funded by NIH/NCRR. The subproject andinvestigator (PI) may have received primary funding from another NIH source,and thus could be represented in other CRISP entries. The institution listed isfor the Center, which is not necessarily the institution for the investigator.We performed a first attempt at micro fluorescence tomography on tumor and normal tissue samples. The setup used the newly developed optics that combines a single-bounce capillary in combination with the main beamline optics. This arrangement delivers 10 times more flux than the current KB mirrors and provides a focal spot of ~10 microns. The samples are normal and tumor tissue samples 60 microns thick. The samples were mounted on an XY stage to generate the tomography data. In the first part of the experiment we excited the sample with 12 keV energy and we collected fluorescence signals from Ca, Fe, Cu and Zn simultaneously. These data will be use to reconstruct the elemental 3D distribution within the samples. The data collected is still under analysis. We found a number of deficiencies in the system that will require some additional effort. This first attempt indicates the feasibility of this kind of experiment at the BioCAT microprobe although the system is not yet optimized.
