This subproject is one of many research subprojects utilizing theresources provided by a Center grant funded by NIH/NCRR. The subproject andinvestigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed isfor the Center, which is not necessarily the institution for the investigator.Plunge freezing methods are used routinely by NRAMM for preserving particulate complexes in a thin layer of frozen buffer. The temperature of the liquid ethane is a critical factor in obtaining good quality vitreous ice. The ethane is kept cold by a surrounding bath of liquid nitrogen which has a lower boiling point (-195.8C) than the melting point of liquid ethane (-182.8C). Consequently, the ethane will freeze solid unless the ethane is regularly heated. This is done by manually stirring the ethane at regular intervals which may heat the ethane well past its melting point which is likely to affect the quality of the ice. The objectives of this project are to design and construct a device that will automatically maintain the liquid temperature at a constant temperature.
