The Transgenic Animal Model Core will be used by approximately 11 UM-MAC investigators who are pursuing gene regulation and expression projects. Specific applications include: (1) Preparation of Transgenic Mice: The core will carry out all the steps necessary to produce transgenic mice from cloned DNA provided by the investigator. Gene constructs will be microinjected into the pronuclei of fertilized mouse eggs. Enough embryos will be injected to produce at least 3 transgenic mice. The eggs will be surgically transferred to pseudopregnant female mice, and after the mice are born they will be transferred to MAC investigators who will screen for the presence of the transgene and carry out breeding regimes appropriate for their project. Transgenic mice can be used for many applications, including the study of tissue specific gene expression, production of animal models of human disease, and characterization of functional effects of introduced mutations. (2) Training: MAC investigators who need many DNA constructs to complete their work will be trained in the preparation of transgenic mice. Training will also be available for other aspects of mouse embryology, including construction of aggregate chimeras and studies of early mouse embryos. (3) Transgenic Support: Support will be provided to UM-MAC investigators who are preparing transgenic mice or conducting studies on early mouse embryos. This support will include quality tested media for embryo culture, hormones for the superovulation of mice, miscellaneous glass and plastic supplies and dissecting instruments, equipment maintenance, and access to the specialized microscopes, micromanipulators, micropipet puller, and microforge necessary for the preparation of transgenic mice. We expect to prepare an increasing number of transgenic mice for MAC investigators during each year of the next cycle. The core is equipped to handle up to 40 constructs per year for investigators throughout the University. The MAC support will subsidize approximately 35 constructs for MAC investigators during the 5 year grant period.
