Older reproductive aged women have higher estradiol levels across the follicular phase, but it is unclear whether this is secondary to increases in FSH or to intrinsic changes in aromatase activity in the ovary that occur with ovarian aging. The purpose of this protocol is to determine whether older reproductive aged women have an increased estradiol response to a fixed amount of FSH both early in the cycle and in the late follicular phase. We have not initiated the early follicular phase portion of the study at this time and therefore this report pertains only to the late follicular phase studies. The late follicular phase protocol involves daily blood sampling from day 1 of the menstrual cycle for a maximum of 18 days. Ultrasounds occur between days 1-4 and 7-10 and then as indicated until the dominant follicle reaches 16 mm or greater. At that time, subjects receive a single subcutaneous injection of rhFSH (Gonal F/Follistim) and blood sampling continues every 6 hr for 36 hr. At 48 hr there is a final blood draw and a final ultrasound.
