A HPV transgenic model with clear and reproducible histopathologic progression of cutaneous lesions is being used to study the formation of squamous cell carcinomas. Genomic alterations are being characterized in histopathologic lesions from HPV transgenic mice. The CGH pattern is being determined in invasive tumors and dysplastic lesions adjacent to these tumors. There are specific regions of the genome that display loss or amplification of specific regions of the genome in a high percentage of lesions. Any regions which give a consistent alteration in pattern are examined employing FISH analysis. Advanced anal lesions (papillomas or advanced dysplasias) are being examined for CGH. The effects DFMO on the development of histopathologic lesions in HPV transgenic mice is being examined. DFMO is being administered in the drinking water. The mice are being examined for the development of lesions on the ears or on the trunk for CGH, FISH, BUDR labeled cell division, and histopathology.
