This subproject is one of many research subprojects utilizing theresources provided by a Center grant funded by NIH/NCRR. The subproject andinvestigator (PI) may have received primary funding from another NIH source,and thus could be represented in other CRISP entries. The institution listed isfor the Center, which is not necessarily the institution for the investigator.The purpose of this study is to demonstrate that triclosan inhibits inflammation and tissue destruction of the oral cavity. A total of 12 participants will be recruited for the study. Subjects will attend only one study visit. They must not show any signs of periodontitis to participate in the study. Forty (40) ml of whole blood will be collected by venipuncture. In order to study inflammation, various experiments will consist of the evaluation of the inflammatory response that will be determined ex vivo using whole human blood that will be stimulated to mimic the human in vivo response. For the in vitro part of the study, the THP-1 cells will be stimulated simultaneously withEscherichia coli LPS and Triclosan. While in parallel, cultures of gingival fibroblast cells will be stimulated with Escherichia coli LPS and Triclosan. Both parts of the study will test triclosan using a solution of 1 part toothpaste, 2 parts water and 1 part saliva to determine the effect of triclosan on thecells. Cell lysates will be collected at 2, 6 and 24 hour time points. At 2 and 6 hours, mRNA extractions will be performed to provide a set of early and immediate response genes. At 24 hours the sample will becentrifuged and utilized for inflammatory mediator analyses. Two control samples not treated with Triclosan will be included.
