This subproject is one of many research subprojects utilizing theresources provided by a Center grant funded by NIH/NCRR. The subproject andinvestigator (PI) may have received primary funding from another NIH source,and thus could be represented in other CRISP entries. The institution listed isfor the Center, which is not necessarily the institution for the investigator.The goal of the protocol is to systematically evaluate the role of genes in the accumulation of drugs into human milk. By identifying which transporters are present in mammary epithelial cells we hope to establish not only the identity of transporters for nitrofurantoin and cimetidine, but to focus future studies on those classes of drugs likely to accumulate into milk. Functional studies will substantiate the role of these expressed transporters in the specific accumulation of nitrofurantoin and cimetidine. The further development of a model cell system that mimics the human mammary epithelial cell barrier will significantly enhance our understanding of the mechanisms of drug transport into milk.
