The laboratory core provides centralized resources for recombinant protein production, protein purification and standard quality control of these regents for the individual projects. This is an existing core unit that has established all aspects of transfection, selection and large scale growth of mammalian cell lines for the recombinant production of Vit.K dependent coagulation factors VIIa and X. Expression of soluble TF and site specific mutants in E. coli or S. cerevisiae are established routine procedures, including refolding and purification to homogeneity for successful crystallization of these proteins [1,2]. Protocols for the purification of plasma proteins, as well as, for the antibody production and fragmentation are in place. This core thus centralizes the production of essential reagents used by the various projects.
