The Phenotype Core has continued to thrive as a facility for support of P&F investigators and others in thelocal, national and international research communities, based on its sound reputation for skin research with aparticular focus on tissue repair. There are two components to the Core: 1) a cell culture laboratory thatprovides cell and adenovirus production facilities to Vanderbilt clients; and, 2) a surgical model facility thatexecutes a variety of animal models for wound repair and provides biomechanical, biochemical, andhistomorphometric data to users. The surgical facility is also extensively used for training of otherinvestigators in quantitative methods of wound healing analysis.Core resources include a fully equipped, dedicated small animal surgery with two surgical stations, gasanesthesia, a video-equipped operating microscope, and laser Doppler imaging of blood flow in situ. Themain laboratory houses a classic Instron tensiometer for incisional wound evaluation as well as a BTC 2000tensiometer for in situ biomechanical measurements on skin and wounds. DNA and protein content arehandled by routine, multi-well spectrophotometry, while collagen analysis is based on highly sensitive HPLCquantification of hydroxyproline and other amino acids from hydrolyzed tissue homogenates. Image analysisutilizes both high-resolution, low magnification digital scanning for archival documentation as well as a stateof-the-art Olympus DP71 CCD coupled to an Olympus BX50 photomicroscope. Fluorescence microscopywith an older Nikon 200 has also been added to the facility. Typical morphometric evaluation of woundsincludes the determination of degree of epithelization, cross-sectional area and height of granulation tissue,and degree of wound closure (gap between intact dermis at the wound margins). Image Pro and Image Jsoftware acquire morphometric data for export into Excel with an added Analys-it package for completestatistical interpretation. The Core also maintains a facility for utilization of the Helios Gene Gun, a device forbiolistic propulsion of DNA-coated gold particles in to cells and tissues.
