ABSTRACT: The use of the Tecnai F20 has increased to about 35 hrs/week (1830 hours during this reporting period of 4/1/05-3/31/06). Some of this increase was due to the 418 hours down time of the F30, which forced users, including those doing tomography, to work at lower voltage. Training of four new users was performed on this microscope with great success. We had 94 hours of service, development, and training. Figure 1 illustrates F20 usage. Plastic section tomography (single and dual axis), closely followed by regular EM work (pre-screening of grids before imaging on the Tecnai F30, negative stain samples, serial section work, single-particle work, and immuno-EM) comprised the largest use of the F20, followed by montage tomography, cryo tomography, service, development (hardware and software) and training of new users, and finally cryo imaging (single particle, evaluation of cryo samples). Figure 2 illustrates the distribution of tomography work done (a total of 837 tilt series taken).
