This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. High resolution data (2.0-2.2 for variant Cre recombinase-Lox DNA complexes will be collected for structure refinement. These complexes test hypotheses concerning i) how Cre recognizes its natural DNA substrate LoxP at different points in the reaction cycle, ii) how differential interactions of Cre with mutant DNA substrates alter reactivity and iii) how the substrate may promote the reaction. Moderate resolution data sets (2.7-3.4 have already been collected, demonstrating the usefulness of these crystals. Trial beam time is also requested to evaluate the diffraction of crystals of a mitochondrial DNA packaging protein, ABF2, in complex with 27mer DNA. These crystals presently diffract 3.2 A- 4.5 using CuKa radiation.
