This subproject is one of many research subprojects utilizing theresources provided by a Center grant funded by NIH/NCRR. The subproject andinvestigator (PI) may have received primary funding from another NIH source,and thus could be represented in other CRISP entries. The institution listed isfor the Center, which is not necessarily the institution for the investigator.We are using x-ray diffraction methods to address issues concerning the biochemical mechanisms of several metal-containing proteins. In addition to determining the structures of novel enzymes, our main focus is the structural characterization of mechanistic relevant intermediates of various enzymes with the goal of obtaining information concerning enzyme mechanism. Since these studies are often concerned with the characteriztion of bound substrates and inhibitors, the details of their interaction with the enzymes can only be revealed with high resolution x-ray diffraction data. Currently, this approach is being applied to metal-containing hydrogenases and nitrogenases, various phosphatases, and enzymes involved in the bioconversion of alkenes and ketones.
