This subproject is one of many research subprojects utilizing theresources provided by a Center grant funded by NIH/NCRR. The institution listed isfor the Center, which is not necessarily the institution for the investigator.There is growing evidence showing that electronegative LDL [(LDL(-)], a circulating proatherogenic LDL particle, is relevant to oxidized LDL (oxLDL) by their biological performance. Our group currently is dedicated to the (LDL(-) study. We have prepared our own (LDL(-), named L5, with well-defined characterization based on composition profile, lipoprotein category, biological activity and oxidation resistance. Our previous experiments have indicated that L5 shares the same characteristics of normal LDL in density and size.4 LDL (~2.5 MDa) is a sphere particle about 22 nm in diameter, containing sole apolipoprotein B-100 (~ 550 kDa), phospholipids and cholesterol on the surface, while triglycerides and cholesterol esters in the core. We postulate that L5 could transition from sphere to rod according to its physical properties and chemical composition. By utilizing electron microscope (EM), we will be able to visualize the size and the shape of L5 unambiguously.
