The institution listed isfor the Center, which is not necessarily the institution for the investigator.Toxicity of thiopurines results after being metabolized and incorporated into DNA as 6-thiodeoxyguanosine. The alteration of local DNA structure by thiodeoxyguanosine and/or its metabolite methyl-6-thiodeoxyguanosine alters important DNA/protein interactions including DNA-mismatch repair, topoisomerase II, telomerase, and RNase H. We are using solution NMR spectroscopy to characterize the changes in local DNA structure and dynamics in 6-thioguanine modified oligonucleotides
