This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The project involves the characterization of mutants of BHMT that are designed to alter the oligomerization properties of the enzyme. The goal is to determine if oligomerization is essential for BHMT catalytic activity. CD spectra of purified site-directed mutants showed that each protein had little alteration in secondary structure. A portion of the work involves kinetics of different oxidizing agents affecting both zinc and activity loss. We would like to be sure that the oxidants are not causing gross structural damage and the effect is limited to the active site.
