The institution listed isfor the Center, which is not necessarily the institution for the investigator.Activation of the protooncogene homolog Ras in yeast results in activation of protein kinase A (PKA). No assay currently exists to directly monitor activation of PKA in vivo. We propose to test whether an in vivo FRET assay based on a method developed by Roger Tsien and his colleagues for use in mammalian cells will function in yeast. The availability of a FRET assay for PKA activation would greatly facilitate physiological studies on nutrient signaling in yeast.
