This subproject is one of many research subprojects utilizing theresources provided by a Center grant funded by NIH/NCRR. The subproject andinvestigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed isfor the Center, which is not necessarily the institution for the investigator. We are currently working with Ian MacLeod in John Yates's lab to conduct a biochemical analysis of the C. elegans and human kinetochores. Using a variety of different purification strategies, we are attempting to isolate all of the known C. elegans and human kinetochore proteins and in the process identify novel interacting proteins. While C. elegans is not renowned for being an excellent biochemical system, the combination of purification strategies has allowed us to purify a number of kinetochore proteins. We plan to continue this analysis and ultimately hope to additionally identify phosphorylation sites in these kinetochore proteins.
