This subproject is one of many research subprojects utilizing theresources provided by a Center grant funded by NIH/NCRR. The subproject andinvestigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed isfor the Center, which is not necessarily the institution for the investigator.Cohesin is a chromosomal protein complex that promotes cohesion between replicated sister chromatids. Cohesin is a large proteinaceous ring, consisting largely of antiparallel coiled coil, held closed at interacting ATPase domains. We have previously developed a model for ATP hydrolysis-dependent transport of DNA into the cohesin ring, where sister chromatids may become topologically trapped. We would now like to analyse movements of the ATPase domains and associated subunits during the individual steps of cohesin binding to DNA.
