This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. Primary support for the subproject and the subproject's principal investigator may have been provided by other sources, including other NIH sources. The Total Cost listed for the subproject likely represents the estimated amount of Center infrastructure utilized by the subproject, not direct funding provided by the NCRR grant to the subproject or subproject staff. Dr. Baiola's group is interested in the N-linked glycosylation of RE1-silencing transcription factor (REST) which relates to its being interaction. REST was produced by HEK-293 transfection with rest cDNA and FLAG-tag. Western blotting revealed that REST contains three different forms of the protein, with apparent molecular weights of 220, 150, and 120 kDa. The apparent molecular weight changes from 220 kDa to 150 kDa after enzymatic deglycosylation treatment using EndoF1. In this study, the Azadi group is investigating the site of glycosylation of this glycoprotein.
