This subproject is one of many research subprojects utilizing theresources provided by a Center grant funded by NIH/NCRR. The subproject andinvestigator (PI) may have received primary funding from another NIH source,and thus could be represented in other CRISP entries. The institution listed isfor the Center, which is not necessarily the institution for the investigator.We propose to use a recently developed coupled capillary-electrophoresis / x-ray fluorescence instrument to determine the metal loading in metalloproteins. Preliminary experiments conducted at the NSLS permitted measurements to a detection limit of approximately 80 M. With the higher flux and faster fluorescence detectors available at SSRL, we estimate that this detection limit can be extended to the nanomolar range. In addition, the higher flux should permit measurement of XANES, and perhaps even EXAFS spectra for resolved samples.
