This subproject is one of many research subprojects utilizing theresources provided by a Center grant funded by NIH/NCRR. The subproject andinvestigator (PI) may have received primary funding from another NIH source,and thus could be represented in other CRISP entries. The institution listed isfor the Center, which is not necessarily the institution for the investigator.A spectral analysis capable flow cytometer is being developed. Currently, all flow cytometers separate fluorescence emissions by dichroic mirrors plus spectral filters. The goal of this development is to analyze the fluorescence emissions by recording with a line CCD array spectrally dispersed light. Preliminary calculations indicate that back thinned CCD arrays have sufficient sensitivity to detect spectrally dispersed light from cells that are in the laser beam for 100 microseconds. A back thinned detector evaluation kit has been obtained from Hamamatsu Corporation. It is being integrated into an existing flow cytometer for evaluation. Pending the outcome of the tests, we will purchase s CCD array for permanent integration into the flow system.
