This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The difference between OMTKY3 and its deritives in their hydrolysis constant may reflect their different dynamic properties. By use of high field of 2D heteronucler NMR experiment, The dynamics properties of OMTKY3 and OMIPF3 will be investigated. The experiments will include measuring hte 15N t1,t2 and heteronuclear NOE for virgin forms and modified forms of OMTKY3 and OMIPF3.The origin of alternative conformation for Asp14OMTKY3 at lowpH is also being investigated(including the hydrogen bond difference)
