The institution listed isfor the Center, which is not necessarily the institution for the investigator.To develop and implement MHC typing technologies for Cynomolgus macaques (Macaca fascicularis) and Indian Rhesus macaques (Macaca mulatta). We will adapt technologies from HLA typing in humans for MHC class I and class II typing in the macaque.cDNA libraries were constructed from 13 Indian Rhesus macaques from the Wisconsin National Primate Research Center colony. These animals were completely characterized for their MHC Class I and Class II genes. 54 novel MHC Class I and Class II alleles were identified from these animals (and from 18 cDNA libraries made the previous year). At least two copies of each novel allele were full length sequenced in both directions, properly named and submitted to Genbank and the Immuno Polymorphism Database. Additionally, two stable transfected cell lines were produced that express either Mamu-DRBW*20101 or Mamu-DRBW*2104.83 MHC class I alleles in cynomolgus macaques of Vietnamese, Chinese and Mauritian origins have been defined. Additionally, 29 MHC class II alleles from Mauritian cynomolgus macaques have been defined. A microsatellite map containing more than 30 loci distributed throughout the MHC was developed and used to define Mauritian cynomolgus macaque haplotypes. Seven haplotypes account for all of the MHC diversity in this population. The MHC class I and II alleles on each of the six haplotypes is now known. PCR-SSP assays for more than 10 Mauritian, Indonesian, and Filipino cynomolgus macaque MHC class I alleles were developed.
