The interaction of the pituitary protein neurophysin with the hormones oxytocin and vasopressin will be further characterized through studies aimed at localizing the region of neurophysin participating in binding and in quantitating the thermodynamic parameters associated with the formation of individual noncovalent bonds in the hormone-protein complex. Specific approaches planned include NMR studies of the distance between spin-labels and selected protein proton resonances, studies of the effects of protein modification on binding, X-ray crystallographic studies (collaborative) and studies of the thermodynamics of binding of systematically related small peptide analogs of oxytocin and vasopressin.
