In this proposal the investigator is building on previous work with the c- myb proto-oncogene, particularly with regard to its phosphorylation in a negative regulatory region. Having demonstrated that phosphorylation of c- myb on Ser528 results in promoter specific affects on transcription the investigator outlines experiments to determine the changes in phosphorylation on S528 through the cell cycle and following differentiation. Then experiments will be performed to determine if either p42MAPK and p34cdc2 phosphorylate c-myb in vivo. Next, proteins associated with c-myb will be isolated and identified. Finally, novel phosphorylation sites on c-myb will be identified and their putative roles in c-myb function will be determined.
