The objectives of the proposed research are to determine the ultrastructural relationships and quantities of elements during the process of mineralization in cartilage and in long bones of embryonic chicks and rats. Cryo-techniques which minimize loss and dislocation of diffusible substances and quantitative electron probe analysis will be employed. Particular attention will be given to matrix vesicles. Elemental shifts in the calcium-transporting chorioallantoic membrane will also be analyzed. The role of carbonic anhydrase in the skeleton and in the chorioallantois will be studied. Vital stains will be developed to enhance the contrast of cryosections.
